在過去的幾十年中,微流體裝置正在蓬勃發展以落實端點照護之想法;然而微流體裝置在操作過程中,常出現如氣泡阻塞流道抑或因尺寸效應所造成混合效率低等嚴重問題而導致結果不准確。除此之外,微系統仍須電力來驅動這些昂貴且體積相對龐大的泵(pump)或是其他裝置來啟動液體傳輸(transport)和控制流體。近年來離心型微流體興起,其驅動法不僅操作簡單,系統設置相對容易更可解決前述問題;然而,儘管離心型微流體擁有這些優點,其還是仰賴離心機內的馬達製造離心力來控制液體,往往這些機台擁有不小的身軀,小則如CD播放器,大則如迷你離心機般大小,不僅笨重不方便攜帶,這些機器通常更需約110V甚至更高之電壓來驅動,在

缺電地區不可能使用,更遑論實踐POCT的想法。本篇文獻中,開發出一組離心型微流體晶片(Centrifugal Microfluidic Disc, CMD)裝載磁性殼聚糖微球(Magnetic Chitosan Beads, MCBs)作為蛋白載體,並以手壓風扇提供的離心力驅動液體進行分段進樣,利用ELISA的架構並利用TMB呈色反應,最後再以手機拍照及影像軟體分析的方式來進行抗藥性肺炎鏈球菌抗體的檢測與定量。實驗結果顯示,研究中所使用之手壓式離心型微流體晶片系統具有高度專一性,在緩衝溶液中之抗體檢測線性範圍為30-1000 ng/ml,偵測極限為22.67 ng/ml;而在複雜的全血中因個體

差異及檢體環境影響,檢測線性範圍為30-1000 ng/ml,偵測極限為24.83 ng/ml。綜合上述之結果,我們所開發之手壓式離心型微流體晶片系統可提供低成本、省時、方便操作、不須用電且精準之抗體檢測方式,並有潛力應用在其他分子檢測,期望在不久的將來可實際落實端點照護之想法,提升全球之健康水平。
